Cover Feature: Chemoproteomics for Plasmodium Parasite Drug Target Discovery (ChemBioChem 16/2021)

نویسندگان

چکیده

Understanding drugs’ mechanism of action is integral to successful antimalarial drug development. Recent advances in chemoproteomic methods have created an avenue globally profile targets the Plasmodium parasites that cause malaria. Some these strategies even allow direct measurement living intracellular parasites. The cover feature highlights achievement probing drug–protein interactome intact Plasmodium-infected erythrocyte. Emerging Methods in Chemoproteomics with Relevance to Drug Discovery.

A powerful interplay exists between the recognition of gene families, sensitive techniques in proteomics, and the interrogation of protein function using chemical probes. The most prominent methods, such as affinity capture, activity-based protein profiling and photoaffinity labeling, are extensively reviewed in the literature. Here we briefly review additional methods developed in the past 15 ...

Target assessment for antiparasitic drug discovery.

Drug discovery is a high-risk, expensive and lengthy process taking at least 12 years and costing upwards of US$500 million per drug to reach the clinic. For neglected diseases, the drug discovery process is driven by medical need and guided by pre-defined target product profiles. Assessment and prioritisation of the most promising targets for entry into screening programmes is crucial for maxi...

Drug–Target Kinetics in Drug Discovery

The development of therapies for the treatment of neurological cancer faces a number of major challenges including the synthesis of small molecule agents that can penetrate the blood-brain barrier (BBB). Given the likelihood that in many cases drug exposure will be lower in the CNS than in systemic circulation, it follows that strategies should be employed that can sustain target engagement at ...

Metabolomics in drug target discovery.

Most diseases result in metabolic changes. In many cases, these changes play a causative role in disease progression. By identifying pathological metabolic changes, metabolomics can point to potential new sites for therapeutic intervention. Particularly promising enzymatic targets are those that carry increased flux in the disease state. Definitive assessment of flux requires the use of isotope...

High-throughput Plasmodium falciparum growth assay for malaria drug discovery.

New therapeutic agents for the treatment of malaria, the world's most deadly parasitic disease, are urgently needed. Malaria afflicts 300 to 500 million people and results in 1 to 2 million deaths annually, and more than 85% of all malaria-related mortality involves young children and pregnant women in sub-Saharan Africa. The emergence of multidrug-resistant parasites, especially in Plasmodium ...
